PPLO Broth With Crystal violet
PPLO Broth Base w/ CV (Mycoplasma Broth Base w/ CV) with the addition of enrichment, is used for isolation and cultivation of Mycoplasma species (pleuropneumonia like organisms) from clinical specimens and mixed cultures.

| Composition | Formula per liter of medium: Beef Heart infusion from 250 gm.; Peptic digest of animal tissue 10 gm.; Sodium Chloride 5 gm.; Crystal Violet 0.01 gm.; Final pH 7.8 ± 0.2 at 25°C |
| Preparation | Suspend 21 grams in 700 ml distilled water. Heat if necessary to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Cool to 45°C and aseptically add 2.85 ml of 1% Potassium Tellurite along with 300 ml Horse serum or 10 vials of Mycoplasma Enrichment Supplement. Mix well and dispense into sterile test tubes. 25% Ascitic fluid can be used instead of Horse serum. |
| Storage | BioScien PPLO Broth with CV should be stored between 10-30°C in a firmly closed container and the prepared medium at 2-8°C. Use before expiry date on the label. On opening, product should be properly stored dry, after tightly capping the bottle in order to avoid lump development due to the hygroscopic nature of the product. Improper storage of the product may lead to lump formation. Store in a dry ventilated area protected from extremes of temperature and sources of ignition. Seal the container tightly after use. Product performance is best if used within stated expiry period. |
| Appearance | The color of BioScien PPLO Broth with CV is Cream to yellow homogeneous free flowing powder. Prepared Medium Yellow coloured may have purple tinge, clear solution in tubes. If there are any physical changes for powder or signs of deterioration (shrinking, cracking, or discoloration), and contaminations for hydrated media, discard the medium. |
