Shapton Medium is recommended for enumeration of spores of Bacillus stearothermophilus

| Composition | Formula per liter of medium: Casein enzymic hydrolysate 2.50 gm; Peptic digest of animal tissue 5.00 gm; Yeast extract 1.00 gm; Beef extract 3.00 gm; Dextrose 1.00 gm; Bromo cresol purple 0.025 gm; Agar 15.00 gm; Final pH (at 25oC) 7.4 ± 0.2 ** Formula adjusted, standardized to suit performance parameters. |
| Preparation | Suspend 27.53 grams in 1000 ml distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Mix well and pour into sterile Petri plates. |
| Storage | BioScien Store below 30°C in tightly closed container and the prepared medium at 2-8°C. Use before expiry date on the label. |
| Appearance | The color of BioScien shapton Medium is Light yellow to grey homogeneous, free flowing powder. Prepared Medium, Light purple coloured, clear to slightly opalescent gel forms in Petri plates. If there are any physical changes for powder or signs of deterioration (shrinking, cracking, or discoloration), and contaminations for hydrated media, discard the medium. |
